It is 1943 and the Royal Navy must rescue 2000 British sailors trapped on an island in the Aegean before the Germans can attack. But the pair of giant Nazi guns on the close-by island of Navarone prevents any evacuation attempt. An air strike is out of the question so a band of six Allied commandos is pulled together to infiltrate the island. They are disguised as Greek fishermen and plan to scale the massive cliffs to set explosives to destroy the guns. When leader Major Roy Franklin (Anthony Quayle) is injured expert mountaineer Captain Keith Mallory (Gregory Peck) takes over command. He clashes repeatedly with Corporal Miller (David Niven) who is the team’s explosive expert. Mallory must also deal with Greek sniper Andrea Stavros (Anthony Quinn) who has sworn to kill him in revenge for causing the death of his family. A savage storm, inaccurate intelligence, sabotage and two beautiful women, Maria (Irene Pappas) and Anna (Gia Scala), further complicate the situation.
